研究概要

中胚葉誘導におけるアクチビン/フォリスタチン系の役割を明らかにするために、アクチビンのシグナル伝達系の細胞内外での調節機構を検討した。
1.アクチビンII型受容体と相互作用する分子による制御
アクチビンII型受容体の細胞内ドメインと結合する分子群ARIPを単離してきた。ARIP1は神経シナプスにおいてPDZドメインを介しアクチビンIIA型受容体と、WWドメインを介してSmad2/3と結合するが、さらに、PDZドメインを介してβ-カテニンとも相互作用することが分かった。ARIP1の過剰発現がSmad分子によるアクチビンの細胞内伝達を低下させること及びβ-カテニンの核移行を阻害することから、ARIP1がアクチビン/Smad系とWnt/β-カテニン系のクロストークに関わる分子として機能していることが強く示唆された。一方、1個のPDZドメインを有するARIP2及びARIP3もARIP1と同様、アクチビンIIA型受容体細胞内ドメイン及びβ-カテニンと結合する。また、ARIP2のホモログをESTデーターベース中に検索したところ、ARIP2-TMを得た。これら3種のARIP分子は、同一のPDZドメインを有し、スプライシングバリアントと考えられる。各種変異体の細胞内局在を検討しているが、アクチビン受容体の動態に重要な役割を果たしているものと考えられる。
2.新アクチビン結合蛋白質FLRGによる制御
FLRGは2個のフォリスタチンドメインを有し、アクチビン及びBMPと結合する。FLRGの発現調節機構を検討するために、プロモーター領域の構造解析を行った。2.6kbのプロモーター領域には、GATA、SP1、Smad、NF-kappa Bの転写因子結合部位が複数個存在した。種々のプロモーター欠損コンストラクトをHeLa細胞で解析したところ、近位の700bpで十分な転写活性を示すことが分かった。
