神経組織での低分子量分子シャペロンのストレス応答と生理的意義

研究概要

平成12年度の研究成果を以下に列挙する
1)αBクリスタリンもHsp27と同様に、リン酸化を受けると重合状態に変化が起こり、解離することを明らかにした。また、リン酸化され、解離状態のαBクリスタリンは分子シャペロンとしての活性が低下することを、リン酸化部位のセリン残基をアスパラギン酸に置換した変異蛋白質を発現させた細胞レベル、および変異蛋白質を大腸菌で作製し、in vitroで証明した。
2)細胞中のHsp27は各種ストレスに応答して、重合型がリン酸化され、解離することを我々は見いだし報告しているが、今回このストレスによる解離が、各種の蛋白質リン酸化酵素阻害剤で抑制されることを明らかにした。すなわち、ホルボールエステル(PMA)以外の薬剤(亜砒酸ナトリウム、塩化カドミウム。過酸化水素、や高張ストレス)によるHsp27の解離は、p38MAPAKの阻害剤で完全に阻止された。一方、PMAによる解離は、Protein kinase C(PKC)の阻害剤で阻止された。このことはHsp27のセリン残基のリン酸化酵素として、p38MAPKの下流のMAPKAPK-2/3とPKCのδ型が関与しているとする報告を裏付ける結果であたった。
3)尾部懸垂により誘発させた廃用萎縮ラットヒラメ筋では、αBクリスタリンのSer-59のリン酸化が特異的に高進し、特に筋肉ホモジェネートの不溶性分画で顕著であることを明らかにした。すなわち、αBクリスタリン、とくにそのリン酸化型が筋細胞内の蛋白質品質管理に関与していることが示唆された。
4)U373MGヒトグリオーマ細胞の培養液にプロテアゾーム阻害剤を添加して培養を継続すると、数時間内にαBクリスタリンが不溶性分画に増量しはじめ、Ser-59のリン酸化型が著しく増加すること、細胞をSer-59のリン酸化を認識する抗体で蛍光染色すると、核周辺に濃縮されたいわゆるaggresomeが形成され、リン酸化型αBクリスタリンが、その構成成分の一つであることを見つけた。この結果もαBクリスタリンが細胞内の蛋白質の品質管理に関与していることを示唆している。
