研究概要

先に報告者が発見した内皮細胞の酸化LDL受容体LOX-1の機能、病態生理的意義、さらに診断・治療的な応用の可能性について解析し、動脈硬化のメカニズムの解明と新しい診断・治療法の開発の可能性を探ることを本研究では目的とした。
動脈硬化の重要なリスクファクターである高脂血症のモデルウサギを用いて、動脈硬化巣の形成以前の非常に早期から血管内皮細胞でLOX-1の発現が亢進していることを明らかにし、動脈硬化巣の形成の前段階での内皮細胞の機能変化が重要であり、この段階でのLOX-1の発現がその後の形態的変化において重要な役割を果たしている可能性を示唆した。また、この系を用いてアンギオテンシンII受容体AT1のアンタゴニストであるロサルタンがLOX-1の発現を抑制するとともに、動脈硬化巣の発達を抑制することを示し、LOX-1の発現と動脈硬化の進展の密接な関連を示すとともに、アンギオテンシンIIの動脈硬化進展のメカニズムについても示唆を与えた。
本研究で作成したLOX-1遺伝子トランスジェニックマウス、ノックアウトマウスは解析の途上であるが、今後動脈硬化進展メカニズムの解明に有用と考えられる。
一方、遺伝子組み替えにより作成したLOX-1とLDLの構成成分であるアポBの抗体を用いて、血液中のLOX-1リガンド(酸化LDLに相当する)を検出する系を作成した。前述の高脂血症のモデルウサギを用いて測定を行ったところ正常のウサギに比べて数倍の濃度のLOX-1リガンドが検出され、血液中のLOX-1リガンドが血管内皮細胞のLOX-1に作用するという図式が成り立つことを示すとともに、少なくとも高脂血症下においてはLOX-1リガンドが循環血液中にあることを示した。これは酸化LDLは血管壁内で生じるとする従来の説とは一線を画するものであり、重要な論点を提供するものと考えられる。
