蛋白質間相互作用ドメインへのゲノム的アプローチ

研究概要	我々は網羅的2ハイブリッド解析の成果に基づき、そこから相互ドメインを組織的にマッピングする目的で下記の「2ハイブリッドフットプリント法」(2HF)を考案した。まず標的ORFに一方向性に欠失を導入した変異体集団を活性化ドメイン融合ベクター(prey)上で構築、2ハイブリッド選択を行った選択集団と行わない非選択集団とを得る。続いて両集団全体からpreyのインサートをPCR増幅、電気泳動で展開する。非選択集団からは全長インサート断片以下レーン全体に渡るラダーが得られるが、選択集団からのラダーは結合領域の境界までで途切れる。これが2HFの原理である。 2HFを行う際に最も問題となるのは、如何に簡便に一方向性に欠失変異を持ったDNA集団を得るかである。我々は組み換えを利用する新規のdeletion by recombination(DBR)法を考案した。DBRではまず標的ORFの例えばN末端部位で切断して得た線状プラスミドとその切断点に隣接するベクター由来の部分(活性化ドメインコード領域)のC末端側にランダム配列を付加した断片とを用意する。次に、両者を同時に酵母に形質転換すると、両者が共有する活性化ドメインコード領域でDNA鎖の乗り換えが起こり、更にランダム配列部分が切断点下流のプラスミド上のどこかと組み換わったプラスミド、つまり切断点と組み換え点の間が様々に欠失したプラスミド、を保持した酵母細胞集団が得られるはずである。このような発想の下、我々はモデル系でDBR現象を確認、更に基本反応条件を確立した。更に、DBRを酵母MAPKカスケードのスキャフォールド蛋白質STE5に実際に応用し、STE11、STE7結合部位の2HFマッピングに成功した。これにより2HFのfeasibilityが示され、その技術基盤が確立した。現在、実用化を目指して反応の至適化を進めている。
