真核生物におけるリン酸リレー情報伝達ネットワークの構造分子生物学

研究概要	細胞内情報伝達機構に関する最近の研究から得られた一般的理解は、各種情報伝達蛋白質間の相互作用と蛋白質のリン酸化を介したシグナル調節機構に還元できる。真核細胞においてはシグナル受容体型チロシンキナーゼからMAPキナーゼカスケードを介して核内へ情報が伝達される過程にその典型を見ることができる。一方、原核細胞で明らかにされつつある情報伝達系はいささか趣を異にしており、シグナル受容体型ヒスチジンキナーゼを主体としている。さらに、情報伝達蛋白質間の相互作用はヒスチジン残基とアスパラギン酸残基のリン酸化修飾によるシグナルの伝達・制御に基づいたHis-Aspリン酸リレー系が情報伝達の基本をなしている。当初原核細胞に固有と思われたHis-Aspリン酸リレー系が酵母・カビ・高等植物にも広く存在することが明らかになりつつある。このような背景に基づき、本特定領域研究の目的に沿って、我々はHis-Aspリン酸リレー型情報伝達ネットワークの多元的・普遍的実体を分子生物学的・構造生物学的アプローチを併用して理解することを目的とした研究を推進してきた。対象としては真核生物系(分裂酵母・高等植物)に焦点を絞った。分裂酵母では減数分裂を制御する情報伝達系に、高等植物においては植物ホルモンへの応答的情報伝達系に関与すると思われるHis-Aspリン酸リレー型情報伝達ネットワークをそれそれ具体的生命現象として解析した。それぞれの対象に関して、シグナル受容体型ヒスチジンキナーゼ→His-リン酸転移(HPt)情報伝達因子→Asp-リン酸化型情報伝達因子間での多元的情報伝達ネットワークの分子機構に関する多くの知見を得ることができた。ここでは、その成果の代表として、高等植物にHis-Aspリン酸リレー因子の一つとして解析した転写因子ARRに見られる高等植物型Myb様DNA結合ドメインのNMR構造を決定することに成功した(ARR10にみられるシングルMyb様DNA結合ドメイン(B-motif)のNMR構造)。
