研究概要

多能性造血幹細胞(Pluripotent hemopoietic stem cells:P-HSCs)とストローマ細胞(Mesenchymal stem cells:MSC)間の主要組織適合抗原複合体(Major histocompatibility complex:MHC)拘束性を解析し、次のような結果が得られた。
1)P-HSCとMSC間のMHC拘束性を解析する新しいin vitroの系を確立した。
2)Cobblestone colonies(CC)数は両者間のMHC不一致の場合ではMHC一致の場合の30%〜50%であった。
3)MHC Class I分子(マウスではH-2K,D,L)を発現していないβ2-microglobulin knock outマウスのHSC又はMSCではMHCの一致、不一致に関係なく、少量のCCしか形成されなかった。
4)HSCのMHC Class Iに対する抗体でHSCを前処理してから共培養するとCCの数が著明に減少した。
5)MSCのClass Iに対する抗体でMSCを前処理してから共培養するとCCの数が著明に亢進した。
6)この際、SCF,FIt3-L,bFGF,IL-6の発現がMSCで亢進していた(Stem Cells 19:46-58,2001)。
以上のin vitroの事実に基づいて、chimeric-resistantなBMTの系ではdonorのMSCをP-HSCと同時に移植する必要があることが判明し、効率良く両者を移植するための骨髄内骨髄細胞移植(BMT via intra-bone marrow injection:"IBM-BMT")の方法を開発した(Blood in press and Stem Cells 18:453-456,2000)。
