水チャネルの遺伝子発現と機能特性から見た植物多細胞系の構築

研究概要	水チャネルは浸透圧差に依存した水の膜透過を促進する。水チャネルファミリーは動植物に広く分布するが、植物では細胞膜に加えて液胞膜にも存在し、分子種の数が多いのが特徴である。本研究は、多数の水チャネル分子の生理的機能と組織内での分布を明確にする目的で、ダイコンの細胞膜(PAQ)と液胞膜(VM23)の水チャネルに焦点をあて、1)各分子種の遺伝子発現、蓄積量の細胞特異性の解明、2)水ストレスへの応答、3)耐乾燥性植物への水チャネル遺伝子の導入と形質への影響の解析を目的とした。結果の概要は以下の通りである。1)水チャネルは普遍的な膜要素としてどの細胞にも均等に存在するとの推定に反して、明確な細胞特異性が明らかになった。いずれのPAQも幼根・胚軸の中心柱、肥大根の形成層、葉柄の維管束鞘にmRNAとタンパク質の蓄積が認められた。また、VM23の場合は、幼根・胚軸の中心柱、葉柄の維管束鞘に顕著な蓄積がみられ、肥大根では柔組織全体に分布していることが明らかとなった。いずれも成長の盛んな細胞、維管束周辺などの水透過量の多い細胞であり、生理的合目的性に合うと判断した。2)ダイコン実生に0.3Mのマニトールで浸透圧ストレスを与えると、PAQの数種の発現量は2,3時間後に一過的に上昇し、やがて減少した。したがって高浸透圧ストレス初期には水チャネルを増やし、長期的ストレスには水チャネルの発現を抑えて水の細胞外への流出を抑制しているものと推定した。3)CAM植物カランコエへの水チャネル導入に成功し、現在、性質を解析中である。
