アサガオの組織分化に関する変異体の解析

研究概要	本研究の目的は、高等植物の組織分化に関する易変性変異体を材料に、その原因遺伝子を機能解析することで、組織分化過程の一端を明らかにすることである。ここでは1)トランスポゾンが易変性変異だけでなくアサガオの多くの安定な変異座を誘発しているという仮説の検証、2)トランスポゾンにより誘発されたと思われる、アサガオの「松島(yellow-mutable)」とマルバアサガオの「黄緑葉(yellow leaf)」変異座の単離を試みた。これまでに解析した4つの易変性変異座の全てにTpn1ファミリーと命名したトランスポゾンの関与を証明しているが、新たにアサガオの花色に関わる6つの安定な変異座を解析したころ、3つの変異座はTpn1ファミリーに属する新規のトランスポゾンが変異の原因であることを見出した。さらに2つの変異座についてもTpn1ファミリーのトランスポゾンが脱離した際に生じたと思われる小さなDNA再編成の存在を確認した。これは、安定な変異座の多くもトランスポゾンにより誘発されているという仮説を支持する結果てある。さらにマルバアサガオにおいても、Tip100ファミリーに属するトランスポゾンが主要な変異原であることを明らかにしつつある。昨年度までにTpn1とTip100ファミリーの配列を利用した遺伝子同定法、TD(Transposon Display)法を開発ており、「松島」と「黄緑葉」変異座についても原因遺伝子中の単離・同定を試みている。
