蛋白質中電子移動経路を決める新しい方法

研究概要	昨年度までに、タンパク質中のすべての原子対間の電子トンネルカレントを分子軌道法で求め、大きなトンネルカレントを持つ原子対をつなぐことによって、電子移動経路を表現した。今年度は微視的な原子間トンネルカレントに中間的な統計平均操作を施すと、電子移動経路の新たな性質が得られることを見出した。具体的には、Ru-modified azurinを用いる。ドナーは本来のazurinが持っているCu+で、アクセプターは部位特異的に置換したHisに配位したRu3+である。このazurinを300Kの熱揺らぎに晒せて、さまざまな構造を集める。各構造毎にトンネルカレントを書き、電子トンネル因子\|T\|を計算する。そうすると、\|T\|は2桁の揺らぎを示した。電子移動速度は\|T\|の2乗に比例するので、4桁の揺らぎを示すことになる。異常に大きな揺らぎである。これは6箇所の部位特異的に置換したHisに配位するRu3+アクセプターにすべて当てはまった。したがって、ユニバーサルな性質である。その原因をしらべると、大きな\|T\|を持つときにはトンネルカレントの向きが揃って、スムースな流れにまっている。逆に、\|T\|が小さいときには、トンネルカレントがスムースに流れているとはいえない。定量化するために、平均的にカレントが行きつ戻りつする回数を指標Qで表し、destructive interferenceの程度を表現した。さまざまな蛋白構造で求めた\|T\|とQの相関を求めると、\|T\|はQに逆比例の関係があることがわかった。これから、Qを調節することによって、電子移動速度を最大4桁程度制御する道が開けた。
