研究概要

アジサイの花色変異機構の解明
19世紀以来の課題であったアジサイの生きた花の発色と変異機構を、はじめて化学的に明らかにした。
本研究では、萼片から有色プロトプラストを得る方法を開発し、一色素細胞でのpH測定と色素成分の超微量分析に成功し、細胞内での発色条件を決定し、色再構築実験によって一細胞レベルでの発色-組成-pHの相関を決定し、赤色と青色アジサイの発色の仕組みを明らかにできた。
アジサイの色素成分を萼片から抽出、単離し、アントシアニンとして、デルフィニジン-3-グルコシドシド(Dp3-G)、コピグメントとして、イソクロロゲン酸(5CQ)、P-クマロイルキナ酸(5pCQ)、クロロゲン酸(3CQ)を得た。金属分析で得た結果をもとに色再構築実験から中心金属がアルミニウムイオンであることを決定した。また、発色とアントシアニンと共存する助色素のキナ酸誘導体や金属イオンの組成比とpHの関係を色再現実験によって明らかにした。青色発色には、pHが3.8で5CQ、5pCQとAl3+が必須であること、赤色では、pH3.5以下であることが条件であった。
個々の細胞は、プロトプラスト化して、それぞれの有色細胞をマイクロマニュピュレーターを用いて分離した。生きた一細胞の吸収スペクトルは、顕微分光法によって測定し、同細胞を新に開発した細胞内イオン電極でpH測定した。新に開発したミクロHPLCによって、一細胞で色素成分分析に成功した。また、Al3+は同色の30個を用いて分析した。青色アジサイ発色は、液胞pHが3.9で、成分組成比、Dp3-G:5CQ:5pCQ:3CQ:Al3+=1:7:3:9:1であり、赤色ではpHが3.2で、比は、1:2:1:16:0.01であった。これらは、in vitroでの再構成実験で再現できた。ここに、はじめて、アジサイの発色と変異機構を明らかにすることができた。
