コレステロール生合成の鍵酵素となるフラビン酸素添加酵素に関する生物有機化学的研究

研究概要	ラット組み換え酵素を用いて、天然より酵素阻害剤のスクリーニングを行ったところ、これまでコレステロール低下作用が認められていた緑茶のカテキンEGCG(IC_<50>=0.69μM)や、生薬大黄由来のガロイルグルコースなどの没食子酸エステルに、強く選択的な阻害活性が認められた。ガロイル基の欠如した化合物はほとんど活性を示さず、従って活性の発現にはガロイル基の存在が不可欠であると考えられた。次に、鎖長の異なる一連のアルキルガレートについて阻害活性を検討したところ、アルキル鎖の長さに応じて強い活性が認められ、ドデシルガレート(DG)にEGCGを上回る最強の活性が認められた(IC_<50>=0.061μM)。これまで報告されている哺乳動物由来SE酵素の選択的阻害剤と比べても、これらガレートの酵素阻害活性はかなり強い部類に属する。また、さらに阻害活性の最適化をめざし、基質に類似したイソプレン側鎖やフェニルアルキル側鎖をもった一連のガレートを合成して活性を検討したところ、これら化合物も同様に強い活性を示した。SEによる酸素添加反応は、酵素活性中心におけるフラビンC(4a)-O-O-Hの生成を経て進行するものと考えられており、こうした阻害効果は没食子酸エステルの酵素活性中心への結合とガロイル基による活性酸素種のトラッピングによるものと推測された。既に結晶構造の報告され、SEとはアミノ酸レベルで約20%の相同性を示すシュードモナス由来のp-ヒドロキシ安息香酸水酸化酵素の結晶構造をもとに分子モデリングによる検討を行ったところ、実際に酵素活性中心のフラビン結合部位近傍にEGCGに特異的な結合サイトの存在が示唆された。最後に、これらガレートにはSE阻害活性以外にも、がん細胞におけるアポトーシス誘導活性が認められた。
