研究概要

微生物代謝産物から、デュアルスペシフィシティーホスファターゼ(DSPase)VHRに対する阻害剤のスクリーニングを行った。その結果、Neosartorya属の一菌株より産生される4-isoavenaciolide(4iA)が、強力なVHR阻害活性を示すことを見出した。Avenaciolideおよび4iAは抗カビ活性物質として、すでに報告されていたが、その詳細な作用機構解析や標的分子との相互作用に関する報告は無い。そこで我々は今回単離した4iAと標的蛋白質であるVHRとの相互作用について詳細な検討を加えた。
4iAの阻害活性は、VHRとプレインキュベーションする時間に依存していること、インキュベーション後透析しても阻害活性が持続することから、4iAはVHRとMichael付加によって共有結合し、阻害活性を示すことが示された。LC/MSを用いた解析により、IC_<50>値(1.1μM)'付近で4iAはVHR一分子に対し、二分子結合していることが明らかになった。さらにLC/MS/MSにより開裂パターンを検討した結果、結合部位の一つはVHRの活性中心に存在するCys124であり、もう一つは酵素表面のアルファヘリックスに存在するCys171でことが明らかになった。種々のホスファターゼを用いて特異性の検討を行ったところ、VHRと同じDSPaseであるCdc25Bの活性は強く阻害したが(IC_<50>=9.4μM)、Ser/Thr phosphataseであるPP1,PP2Aの活性は阻害できなかった(IC_<50>>250μM)。以上の結果から4iAは、DSPaseの活性中心に共通に存在するCXXXXXRモチーフのCを直接修飾することによりVHRを阻害していると考えられる。
