TH2細胞優位に発現される新規サイトカインレセプターの解析

研究概要

申請者はレトロウイルスベクターを利用した新しいシグナルシークエンストラップ法(Kojima and Kitamura,Nature Biotechnol.1999)を利用してTh1細胞およびTh2細胞に発現されている膜蛋白質あるいは分泌蛋白質をスクリーニングした。スクリーニングにおいてTh2優位の発現を示す新規サイトカインレセプターδ1を同定した(Fujio et al.,Blood 2000)。
δ1は共通γ鎖と相同性を有し細胞内部位にはシグナル伝達に必要なbox1およびbox2領域がある。box1領域はJak2に結合していること、エリスロポイエチンレセプターのbox1と交換すると、エリスロポイエチンの増殖シグナルを伝えることが判明した。その後アメリカNIHおよびImmunex社のグループによりδ-1がTSLPというサイトカインのレセプターの一部であることが報告された。申請者らは現在、δ-1が他のレセプターのサブユニットでもある可能性も考え、ノックアウトマウスを作製するなどさらに詳細な解析を進めている。
