Th1反応における情報伝達制御機構の解明-IL-12Rを介したIFN-γ産生へのシグナル伝達-

研究概要

ヒトIL-12Rβ1およびβ2細胞内領域をBaitとしLexAのTwo-Hybrid法でスクリーニングを試みたがポジティブクローンが得られなかった。そこで、マウスIL-12Rβ1およびβ2細胞内領域をBaitとしGAL4のTwo-Hybrid法を用いマウスT細胞リンパ腫瘍株(V13)由来cDNAライブラリーをスクリーニングしたところ、未知の分子を含む複数個のポジティブクローンが得られた。現在、得られて遺伝子を培養細胞株へトランスフェクトし、IL-12Rβ1およびβ2への会合やIL-12Rを介したシグナル伝達における役割を検討中である。
レトロウイルスベクターを用いた発現クローニングでは、IFN-γのGenomic遺伝子のATGの後にピューロマイシン抵抗性遺伝子を挿入したプラスミドを構築しトランスフェクタントを作製したが、薬剤耐性による選択でIFN-γ発現誘導が識別できなかった。そこで、GFP遺伝子を挿入したプラスミドを構築しT細胞株のトランスフェクタントを作製し、IFN-γ発現誘導をGFPの蛍光強度の増大で見られる系を確立した。現在、レトロウイルスライブラリー感染後、蛍光強度が増大する細胞をFACSにより分集し導入された遺伝子を解析中である。
B7-1/2のT細胞側のCounterpartであるCD28/CTLA-4のIL-12R発現増強への関与を解析した結果、機能的IL-12Rの発現にはCD28からの刺激はポジティブにCTLA-4からの刺激はネガティブに働くこと、さらにIFN-γKOマウスを用いても同様な効果が見られることを示し、IL-12R発現におけるB7-1/2-CD28/CTLA-4刺激の重要性を明らかにした。さらに、CTLA-4からの刺激はナイーブT細胞のTh1への分化を増強すること、この際IL-12の関与は少なくTGF-βを部分的に介しいることも明らかにした。
