研究概要

平野はGp130のSHP2結合サイトに点突然変異を導入した変異gp130を発現しているノックインマウスに自然発症する慢性関節リウマチ様自己免疫疾患の免疫学的異常を明らかにした。とくに、胸腺と抹消におけるセルフトレランスの維持が低下していることを明らかにした。胸腺やTリンパ球におけるシグナル異常を明らかにすることで、自己免疫疾患の発症機構を明らかにする基礎を築いた。吉村はサイトカイン制御分子SOCSファミリーのうちSOCS3/CIS3の発現が炎症性腸疾患やリューマチ患部で亢進していることを見いだした。また、SOCS-3アデノウイルスを自己免疫性関節炎モデルに投与したところ劇的な改善効果があることを明らかにした。SOCS3は抗炎症性遺伝子として作用することを明かにした。瀧は、転写因子IRF-2を欠損する炎症性皮膚疾患を自然発症するモデルマウスを使用して、IRF-2はT細胞以外の細胞に必要であることをしめした。すなわち、CD8+T細胞は発症に必須であるが、IRF-2は、T細胞ではなく皮膚細胞で機能していると考えられる。高津はIL-5とCD38依存性のIgHクラススイッチ組み替えにNF-kBとSTAT5の活性化が必須であること、IL-5Rα鎖の細胞膜ドメインのカルボキシ末端がIgHクラススイッチ組み替えに必須であること、さらにアダプター分子、LnkはLin-c-Kit+Sca1+は造血前駆細胞の増殖を負に制御していることを明らかにした。竹田はTLR7が抗ウィルス薬として臨床応用されている合成化合物の認識に必須であることや、TLRを介したシグナルが、自然免疫系の活性化だけでなく、獲得免疫系の活性(Th1/Th2反応のバランス)を制御していることを明らかにした。熊ノ郷は膜型のセマフォリン分子であるCD100がリンパ球の活性化により細胞表面上でproteolyticに切断を受け分泌型の分子として機能する事、自己免疫疾患モデルマウスであるMRL/lPrマウスの血清中における自己抗体価の上昇と一致して分泌型のCD100が上昇する事、更に分泌型のCD100を過剰に産生するtransgenicマウスでは免疫応答が亢進している事より分泌型のCD100が実際の生体の免疫応答において機能することを明らかにした。宮坂は、L-,P-セレクチンはGlcAβ1/IdoAα1-3GalNAc(4,6-O-disulfate)の繰り返し構造を硫酸基依存的に認識することを明らかにした。セレクチンの糖鎖構造には特定のケモカイン、すなわちSLC, IP-10,SDF-1βなどが結合し、この結合も糖鎖構造の硫酸基に依存的であった。以上の結果は、セレクチン、ケモカインの間のクロストークを媒介するものとしてグリコサミノグリカン(GAG)鎖が重要であることを明らかにしたものである。
