T細胞の副刺激情報伝達の解析と情報変換によるアポトーシス誘導

研究概要	我々は昨年、種々の副刺激分子がraftに存在することと、それらの分子に対する抗体と抗CD3抗体の共架橋によりTCR signalingに重要なraftとTCRの集合が促進され、TCRシグナルが増強することを見い出し、これが副刺激のメカニズムであることを示した。一方、CD28はraftに存在しないにもかかわらず、raftを集合させることが分かった。T細胞表面分子はAPCとの結合時に接着面に移動し、この移動にCD28シグナルが関与することが報告されている。そこで、CD28がT-APC接着およびraftの凝集に与えるシグナルを解析し次の成果を得た。(1)抗原特異的なT-APC結合の接着面にraftの凝集が、同時にCD3、CD4、LAT、CD28、Rac、PKCθ、F-actin等の凝集が共焦点レーザー顕微鏡により観察された。(2)抗原特異的なT-APC結合は、抗CD80/86抗体又はRho family GTPase阻害剤であるC.difficle Toxin Bによって阻害され、逆にCD28分子の刺激によって増強された。さらに、(3)抗CD80/86抗体、又はC.difficle Toxin Bによって細胞接着の抑制と共に、各分子及びraftの凝集が阻害された。(4)PAKのGTP結合部位を導入し、内在性のRho family GTPaseの活性を抑制させると、raftの凝集及び膜分子の集合が抑制された。以上より、T-APC接着およびraftの凝集には、CD28由来シグナルが必須であり、Rac、PAKの活性化に基づくことが示唆された。
