ウイルス感染応答のシグナル伝達機構

研究概要	植物がウイルスを認識してから、抵抗性反応が誘導されるまでの間のシグナル伝達系については明らかになっていない。本研究では、病原ウイルスとしてCMVとTMV、宿主植物としてシロイヌナズナとタバコを用いて過敏感反応死の機構とウイルス抵抗性の発現に至るシグナルカスケードを解析した。 (1)昨年の研究から、CMV抵抗性遺伝子RCY1は、シロイヌナズナecotype C24の第5染色体の長腕に存在するマーカーDFRとT43968の間(約400kb)に存在することが明らかになっている。そこで、RCY1についてさらに詳細なマッピングを行うとともに、その領域部分の塩基配列の決定とCMV罹病性ecotype Colにおける同領域塩基配列との比較解析を行った。その結果、抵抗性遺伝子産物に認められるNBSドメインとLRRドメインを含むタンパク質をコードしている遺伝子が見出された。この遺伝子を、Colに形質転換したところ、CMV抵抗性を示したことから、同遺伝子がRCY1であると判断した。 (2)TMV感染により同調的に過敏感細胞死を誘導する系を用い、細胞死誘導のシグナル伝達におけるCa-カルモジュリン(CaM)の役割について解析を行った。TMV感染を受けたタバコより13種類のCaM遺伝子を単離し、過敏感反応や傷害誘導などに伴うそれら遺伝子の発現を、転写レベルとタンパク質レベルで解析した。その結果、異なる3タイプの発現変動が認められた。それぞれの代表的な遺伝子NtCaM3は活性酸素の発生に関与するNAD+キナーゼを、NtCaM13はNOシンターゼを強く活性化し、NtCaM1は中間的な特性を示した。植物において異なるタイプのCaMが、それぞれ転写レベルおよびタンパク質レベルで独自に制御され、過敏感反応や傷害シグナル伝達に関与していることが示された初めての報告である。
