研究概要	研究代表者(高橋)は、広い宿主範囲を持つことから農業上問題となっているキュウリモザイクウイルス(CMV)とシロイヌナズナを用いて抵抗性発現のシグナル伝達系の解析を行った。その結果、(1)抵抗性は植物の単一優性遺伝子[CMV抵抗性遺伝子,RCY1; Resistance to CMV(Y)]により決定されている、(2)RCY1は、分子量104kDaでCC)_NBS_LRR構造を持つタンパク質をコードしている、(3)RCY1遺伝子座は、べと病菌抵抗性遺伝子RPP8やTCV抵抗性遺伝子HRTと対立遺伝子であったことから、それらは、共通の祖先抵抗性遺伝子に由来するものと考えられる、(4)RCY1によるCMV抵抗性の発現には、サリチル酸(SA)とエチレンを介したシグナル伝達系が関与しているが、それら以外に抵抗性発現を調節している新規のシグナル伝達系が存在することが明らかになった。また、研究分担者(大橋)は、タバコモザイクウイルス(TMV)感染タバコにおける同調的HR誘導系を用いてウイルス感染応答におけるシグナル伝達系の解析を行った。その結果、(1)ジャスモン酸(JA)やSAなどのシグナル物質の他、スペルミンはSAとは無関係にこれら防御遺伝子発現やTMV抵抗性を誘導することから、スペルミンを介した新規シグナル伝達系が存在する、(2)新規ペルオキシダーゼ遺伝子を2つ単離し、その発現誘導を詳細に検討したところ、HRシグナル伝達に未知の系が少なくとも2つはある、(3)同調的HR誘導において急速に発現が低下するDS9遺伝子はATPaseおよび亜鉛依存的なプロテアーゼ活性を示すタンパク質をコードしており、このタンパク質の減少が葉緑体の機能を喪失させ、細胞死を促進させる、(4)TMV感染あるいは傷害をうけたタバコより13種類のCaM遺伝子NtCaM1-13を単離し、それらの遺伝子が、TMV感染によるHR、傷害、SAおよびJA処理などにより異なる制御を受けることが明らかになった。
