研究概要

本研究課題では、キュウリモザイクウイルス(CMV)-シロイヌナズナ系とタバコモザイクウイルス(TMV)-タバコ系を用いてウイルス抵抗性のシグナル伝達機構について解析を行った。キュウリモザイクウイルス(CMV)-シロイヌナズナ系では、(1)CMV抵抗性において、サリチル酸(SA)シグナル伝達系とジャスモン酸(JA)シグナル伝達系がnegativeにcross-talkしている、(2)多くの病害抵抗性のシグナル伝達系に関与していることが知られているNDR1とEDS1は、CMV抵抗性シグナル伝達系には関与していない、(3)CMV抵抗性発現には、新たに脂質により調節されるシグナル伝達系が関与していることが明らかになった。さらに、(4)シロイヌナズナcDNA約9,000クローンをスポットしたマクロアレーを用いて遺伝子発現の網羅的解析を行い、抵抗性特異的に発現が変動する遺伝子群を同定した。一方、タバコモザイクウイルス(TMV)-タバコ系では、(1)病害抵抗性シグナル伝達において重要な役割を果たしているMAPキナーゼ(WIPK)の活性化を誘導する新規物質WAF-1を単離し、構造を決定した。さらに、(2)このMAPキナーゼを脱リン酸化する酵素(MAPK phosphatase : MKP)が、カルモジュリン(CaM)と特異的に結合することによって活性が制御されていることが明らかになった。これらの結果は、ウイルス抵抗性の分子機構を明らかにする上で重要な知見を提供すると考えられる。
