エリシターの受容とオキシダティブバーストの分子機構とその機能

研究概要	本年度の研究は、エリシターの受容の分子機構およびその受容に伴い活性化するキシダティブバースト(OXB)系の分子基盤と制御機構と機能の解明を目的とした研究課題の中で、次の研究成果を挙げた。1)キチンエリシター受容体からの情報発信の機構におけるエリシター結合直後に起こる受容体分子の挙動の変化を、高活性キチンオリゴ糖エリシターから合成した様々な親和性試薬を利用した親和性標識実験により調べ、イネ培養細胞原形質膜に存在するエリシター結合タンパク質(受容体候補分子)がエリシターの結合によって膜上で2量体を形成することを示唆した。2)この2量体がエリシター結合タンパク質2分子からなるホモ2量体であることも示唆し、キチンオリゴ糖エリシター受容体の性格および受容体活性化機構に関して重要な示唆を与えた。3)エリシター認識に伴うOXB系の分子基盤の解明の一環として、ジャガイモに次いで、タバコ植物(Nicotiana benthamiana)のgp91phoxホモログとしてNtrbohAとNtrbohBの二種のcDNAをクローニングし、前者は常時発現、後者はエリシター誘導性であることを明らかにした。4)タバコ植物で、それぞれの特異的なN末端領域および共通性の高いC末端領域をPVXベクターに組み込んだものを接種し、ジーンシレンシングを行った結果、それぞれの遺伝子の発現低下を確認し、非親和性疫病菌に対する感染抵抗性と感染およびエリシターに対するOXB応答が低下することを示し、OXBが局部的防御応答に決定的な機能をもつことを示唆した。5)局部的OXBの誘導が全身的シグナル発進と伝達を制御し、全身OXBと全身獲得抵抗性を誘導する機構の解明の一環として、OXBの発生に伴い二次的全身シグナル発進因子が存在し、サリチル酸やBTHがその発進作用をもち、細胞間情報伝達はカルシウムのインフラックスの連鎖反応により起こることを示した。
