植物表層における病原菌-植物シグナル交換と情報伝達

研究概要

植物と病原菌の相互認識は、両者が接触した直後から開始され、それぞれの応答の結果、受容や拒絶が決定されると考えられる。本年度は、植物と病原菌で起こる極初期の認識と応答の実相について解析を進め以下のような知見を得ることができた。
1)病原菌の感染器官形成に関わる遺伝子発現を指標に、情報伝達系を解析した結果、酵母のフェロモン応答性MAPキナーゼ経路に相当する経路が植物病原菌の感染に必須であり、さらにMAPキナーゼ制御化にある酵母Ste12転写因子の相同遺伝子CST1のクローニングに植物病原糸状菌では始めて成功した。また、形態分化、病原性・感染特異性決定機構の遺伝学的解明を目的としてタギング変異株のスクリーニングを行い、複数の形態分化異常株、感染能欠損株の取得に成功した。さらに、病原菌の感染誘導物質と植物の感染行動誘導物質の存在を突き止め、現在これらの精製を進めている。
2)植物細胞の最外層を構成する細胞壁には、病原菌のエリシターやサプレッサーを認識するNTPaseが存在することを明らかにした。次に、NTPaseに近接するperoxidase(POX)を探索し、細胞外の局在が予想される5種のPOXのクローニングに成功した。POXによるO_2^-生成は、NTPaseで生じるリン酸とADPによって制御されることを突き止めた。この結果から、病原菌シグナルは植物細胞壁上のNTPaseで認識され、速やかに防御応答(OXB)を誘導すると考察した。さらに、細胞壁のエリシター結合タンパク質を検索した結果、NTPase以外にトマト抵抗性遺伝子Cf9産物のホモログを見い出した。これはPGIP活性を有する事から、ペクチンからポリガラクチュロン酸オリゴマー(内生エリシター)を生成し、これが第2次シグナルとして、細胞内への情報伝達を担うというスキームを考察した。
