研究概要

本研究の目的は、プラスセンスRNAをゲノムに持つウイルスの植物への盛染過程に関与するウイルス因子と植物因子の役割を抵抗性と侵略の両観点から明らかにし、ウイルス病制御法の開発のための基礎情報を得ることである。
Virus induced gene silencing(VIGS)を利用したウイルス感染関連宿主因子の探索研究では、ToMVの感染を抑制する数種の候補遺伝子を明らかにした。その中で、真核生物翻訳伸長因子(eEF1B)について現在解析中である。
RCNMVのRNA1がRNAサイレンシングサプレッサーをコードしていることを明らかにした。さらに、サプレッサーである複製酵素成分タンパク質(p27とp88)は、タンパク質として存在するだけではRNAサイレンシング抑制機能を持たないが、抑制機能発現にはマイナス鎖合成の鋳型となりえるRCNMVRNAを必要とすることを明らかにした。
TMVの細胞間移行機構の研究では、複製酵素タンパク質(Rep)が細胞間移行に関与すること、そして、Repが移行タンパク質(MP)と共局在することを明らかにした。また、MPが原形質連絡に移行していく画分に存在するgermin like proteinについては、VIGSによる機能解析とGFP融合タンパク質を用いてその発現と細胞内局在を現在解析している。
シロイヌナズナを舞台にしたウイルス感染干渉作用研究では、RNA dependent RNA polymeraseの一つをコードするsde1(ecotype Columbia)のT-DNA tagによる破壊株でも干渉作用がみられることから、いわゆるPTGSのpathwayとは異なる機構、ないしは分子種が関与していると想像される。いくつかの遺伝子破壊アラビドプシス変異株を用いて、干渉現象に関与する遺伝子の候補を探索している。
