研究概要	動物の行動を制御する遺伝子が遺伝学的解析により明らかになっている。その中で、我々は遺伝子欠損マウス作製技術を開発し、Fyn欠損マウスの作製と解析を行ってきた。その結果、様々な行動制御にFynが関わっていることが明らかになった。本研究では、このFynを動物行動制御分子と位置付けて、Fynと関連する遺伝子の解析を行い、シナプスにおけるNMDA受容体の制御、新たなガドヘリンファミリー(CNRファミリー)との結合を明らかにしてきた。現在までにCNRファミリーの分子機構を明らかにすることによる、哺乳動物の行動を制御する新たな分子メカニズムの解明を行っている。その結果CNRファミリーは、1)シナプスに存在する膜タンパク質であること、2)ゲノムDNAにおいてB細胞受容体やT細胞受容体に類似した遺伝子クラスターを形成していること、3)遺伝子クラスターにおいてマウスで14種類、ヒトで15種類の可変領域エクソンが存在し、共通領域である不変領域と連結していること、4)不変領域では、少なくとも2種類のスプライシングの違いによる細胞質領域の構造変換をおこさせることが可能となっていること、5)CNRファミリーの第一カドヘリン領域のアミノ酸配列が高く保存されていること、、6)この第一カドヘリン領域とReelinタンパク質とが結合すること、7)このReelin結合領域に対する抗CNR抗体によりReelin依存的なDab1のチロシンリン酸化が阻害されること、8)第一カドヘリン領域は動物種の違いにより系統発生的にアミノ酸配列が高く変化していること、9)第一カドヘリン領域は大脳皮質の発達過程において、アミノ酸配列の変化をともなう突然変異の蓄積がおこることが明らかになった。以上のように、CNRファミリーはFynと脳の形成、シナプス形成に関わることが予想される興味深い遺伝子ファミリーであることが明らかとなり、今後遺伝子欠損マウスの作製により、分子機能と脳機能との関わりあいを明らかにする予定である。
