発生工学を用いた心不全形成におけるgp130の役割の解析

研究概要	近年心不全の病態に酸化ストレスが関与することが知られ、また心筋特異的血管内皮増殖因子(VEGF)欠損マウスにおいて心筋の機能不全が報告されている。本研究においてSTAT3の心不全病態への関与をこれらの観点より検討を行った。新生児ラット心筋細胞を用い、変異STAT3遺伝子導入にはアデノウイルスベクター系を用いた。in vivoの実験に関しては、α-ミオシン重鎖プロモーターを用いた心筋特異的トランスジェニックマウス(TG)を作成した。 LIFによりgp130を刺激すると、培養心筋細胞においてMnSOD mRNAおよび蛋白の発現増強が認められ、虚血再灌流に伴う心筋細胞傷害が軽減された。LIFの前投与がなくても持続活性型STAT3(caSTAT3)の遺伝子導入によりほぼ同程度の効果が得られた。LIFの作用はドミナントネガティブ型STAT3(dnSTAT3)を導入しSTAT3のシグナルを阻害することにより抑制されたが、MAP kinase阻害剤では影響を受けなかった。同様の結果がVEGFの発現においても観察された。次に、マウスにLIFを投与し心臓におけるVEGFの発現を検討したところ、wild-type mouse (WT)ではVEGFの発現増強が認められたものの、dnSTAT3 TGでは発現増強が観察されなかった。これまでにSTAT3が血管新生に直接に関わっているかについては明らかにされていない。そこで、caSTAT3 TGを作成し心筋内の血管密度を検討した。その結果、caSTAT TG心臓においてWTに比して明らかに血管密度が増加すること、併せて、血管内皮特異的蛋白質であるVE-cadherinの発現が増強していることが示された。 STAT3は、腫瘍形成、炎症、組織再生に重要な役割を演じていることが知られているが、本研究結果によりSTAT3を介したシグナルが、活性酸素種による心筋細胞傷害を軽減し、心筋内の血管形成を促進することにより心不全に伴う心筋リモデリングに重要な役割を果たしていると考えられた。
