研究概要	骨量は女性ホルモンであるエストロゲンの影響を受けることが知られている。エストロゲンは細胞内にあるエストロゲンレセプター(ER)に結合する。エストロゲンを受容したエストロゲンレセプター(ER)は、DNAの特異的配列に結合して、下流に存在する標的遺伝子群の転写を制御する。わたしたちはERがエストロゲン存在下、非存在下においてそれぞれ異なったユビキチンリガーゼによってユビキチン化を受け、分解されることを見出した。エストロゲンの結合していないERはHsc70を含むシャペロン複合体によってCHIPと呼ばれるユビキチンリガーゼと結合し、分解される。CHIPは変異型のERを選択的にユビキチン化することから、ERの品質管理に関与しているものと考えられる。一方、エストロゲンの結合したERは新規ユビキチンリガーゼによってユビキチン化されることが明らかとなった。このユビキチンリガーゼはエストロゲン依存的にERと結合し、ユビキチン化することによって分解へと導く。現在、このユビキチンリガーゼの機能を探るため、遺伝子欠損マウスの作成を進めている。エストロゲンはIGF-1の発現を促進することによって骨芽細胞の増殖を促すと考えられている。わたしたちは、ERがエストロゲン依存的にIGF-1受容体に結合し、IGF-1受容体の活性を促進することを見出した。この結果は、エストロゲンによる骨芽細胞の増殖にERとIGF-1受容体の相互作用が直接かかわる可能性を示唆するものである。
