研究概要	Cbfa1ノックアウトマウスの間葉系細胞が骨芽細胞分化のどの段階でブロックされているかを調べるために、その頭蓋冠の細胞をin vitroで培養しその分化能を検索した。Cbfa1を欠損すると骨芽細胞分化能は完全に失われていたが、培養中に多くの細胞が脂肪細胞に自然に分化していき、またBMP-2とともに培養すると軟骨細胞に分化した。したがってノックアウトマウスの頭蓋冠由来間葉系細胞は、骨芽細胞以外の骨格系形成細胞(脂肪細胞、軟骨細胞)に分化しうる多分化能を持った細胞であることがわかった。このことは、これらの間葉系細胞が非常に未熟な分化段階にあること、すなわちCbfa1が骨芽細胞系列への分化決定の初期に関わっていることを示唆している。次に生体内でCbfa1が軟骨細胞の分化にどのように働いているかを明らかにするために、軟骨細胞に特異的に発現するII型コラーゲンプロモーター・エンハンサーを用いたCbfa1トランスジェニックマウス(Tg)を作製した。Cbfa1 Tgでは、軟骨細胞は早期から成熟肥大化し、骨に置き換えられる所謂内軟骨性骨化が促進した。ドミナントネガティブ型(DN)Cbfa1 Tgでは、軟骨細胞の成熟及び内軟骨性骨化が抑制された。したがって、Cbfa1は骨芽細胞分化因子であるとともに軟骨細胞の分化因子であることを証明することができた。Cbfa1 Tgでは、本来永久軟骨になるべき軟骨をも成長軟骨化しており、ほとんどの関節は癒合していた。逆にDN Cbfa1 Tgでは、本来の成長軟骨でも永久軟骨の形質を保持していた。したがって、Cbfa1は成長軟骨と永久軟骨の性格を決定づける上でも重要な働きをしていることを明らかにできた。
