研究概要

1.Cbfa1の破骨細胞分化における機能解析
Cbfa1欠損頭蓋冠由来細胞株に、Cbfa1を導入すると、RANKLは誘導され、OPGは抑制され、Cbfa1はRANKLの誘導、OPGの抑制を介して、破骨細胞の分化に促進的に働いていることが明らかとなった。また、Cbfa1ノックアウトマウスでは、硬骨細胞がみられず、RANKLの発現も著明に低下していた。この破骨細胞の欠損が、RANKLの著減に起因しているかを調べるために、Runx2ノックアウトRANKLトランスジェニックマウスを作製した。このマウスでは、多核のTRAP陽性細胞が石灰化軟骨周辺に出現したが、破骨細胞分化は充分に起きていなかった。すなわち、M-CSF、RANKL.の発現だけでは、破骨細胞分化には充分ではなく、Cbfa1が最終分化した軟骨細胞や骨芽細胞に発現誘導している基質蛋白や可溶性因子が、破骨細胞分化に重要であることが明らかとなった。
2.Cbfa1の軟骨細胞における機能解析
Cbfa1欠損軟骨細胞は、培養中に脂肪分化マーカーを発現し始め、脂肪細胞へと分化した。これは、Cbfa1の導入により阻止され、軟骨細胞の形質は維持された。TGF-β、レチノイン酸、IL-1β、PDGF、bFGF、PTH、IL-11はCbfa1欠損軟骨細胞の脂肪細胞への分化を抑制し、Cbfa1はTGF-βと相乗的にIL-11を誘導した。したがって、Cbfa1は軟骨細胞の形質維持に重要な働きしているとともに、脂肪細胞分化の抑制に関わっていることが明らかとなった。また、Cbfa1は、一部IL-11の誘導を介して脂肪分化を抑制していた。
