c-Srcリン酸化を介する転写因子NF-κBの活性化と骨吸収の制御

研究概要	1.破骨細胞分化におけるJNK/c-Junの役割(in vitro) 破骨細胞分化におけるJNK/c-Junシグナルの関与を検討するために、ドミナントネガティブ(DN)-JNKおよびDN-c-Junを作成し、アデノウイルスベクターに組み込んだ。DN-JNKあるいはDN-c-Junを破骨細胞前駆細胞株RAW264に過剰発現したところ、可溶性RANKLによる破骨細胞分化誘導能が著明に抑制された。一方、コントロールウイルスは影響を及ぼさなかった。 2.DN-c-Junトランスジェニックマウスの解析 JNK/c-Junシグナルのin vivoにおける役割を検証するために、破骨細胞特異的であるTRAPプロモーターを用いて、DN-c-Junを過剰発現するトランスジェニックマウス(TG)を作成した。このTGでは、長軸方向の成長阻害、歯牙の未萌出、海綿骨の著しい増生および破骨細胞の分化障害を認めた。 3.破骨細胞分化におけるNFATの役割 c-Junと協調する転写因子NFAT1をRAW264細胞に過剰発現させると、破骨細胞様細胞の分化が誘導された。このNFAT1の効果は、DN-c-JunあるいはDN-c-Fosの過剰発現により著しく阻害された。以上のデータより、RANKLにより活性化されるJNK/c-Junシグナルは、破骨細胞分化誘導に必須であることが示された。さらにc-Junは、転写因子NFAT1と協調して破骨細胞分化を誘導できることが明らかとなった。この研究結果は、破骨細胞の分化機構の分子メカニズムを明らかにし、骨代謝性疾患の治療法の開発の大きな指針となると期待される。
