イネにおけるプロテインボディへの輸送・蓄積を制御する遺伝的メカニズム

研究概要	1)57H変異体の遺伝学的解析及び変異体の作出・MNU受精卵処理後代約1,000系統についてスクリーニングを行い、新たに9系統の57H変異体を得た(合計45系統)。変異体と野生型系統及び標識遺伝子系統との交配を行い、多くのF1種子を得た。・遺伝分析の結果、新たに、Glup5遺伝子及びglup6遺伝子を同定した。 2)細胞内輸送・集積に関わる酵素タンパク質の同定と解析・glup3変異体種子では野生型と同等のVPEが検出され、かつ、活性も認められたことから、本遺伝子はVPEの活性制御、もしくはVPE以外の限定分解酵素に関わる可能性が示された。・起源の異なる3種のesp2変異体系統の登熟種子について、60kDPDIプローブを用いたノーザン分析を行った。全ての系統で60kD PDIの発現が認められなかったことから、Esp2遺伝子と60kD PDIとの密接な関係が確かめられた。 3)57H突然変異遺伝子の精密度連鎖地図の作成・glup3遺伝子は染色体4のRFLPマーカーR374(短腕から81.9cM)とR2783(同じく83.2cM)に挟まれた領域1.3cM内に存在していることが明らかとなった。 4)57H突然変異体の免疫組織化学的解析・グルテリン前駆体の局在を蛍光顕微鏡を用いて調べた。esp2変異体の前駆体は0.2ミクロン程度の顆粒にプロラミンと共に存在し、Glup1変異体では0.1ミクロン以下の極めて小さい顆粒にやはりプロラミンと共に存在していた。・glup4変異体の登熟種子の電子顕微鏡観察の結果、glup4変異体のグルテリン前駆体は、esp2変異体と同様に、プロラミンと共にER内に混在し集積されていた。
