G1期におけるDNA複製因子の発現と活性の制御機構の研究

研究概要

転写因子E2Fは、癌抑制遺伝子産物pRbの主な標的であり、細胞周期進行特にS期(DNA複製)の誘導に中心的な役割を果たす。そこでDNA複製誘導におけるE2Fの役割を調べるために、E2Fの新規標的遺伝子を探索している。このために、哺乳動物細胞を用いた系とアフリカツメガエル初期胚を用いた系を進行させている。哺乳動物細胞を用いた系では、休止期のラット繊維芽細胞株REF52にレコンビナントアデノウイルスを用いてヒトE2F1を過剰発現し、サブトラクション法を用いてE2F1過剰発現細胞に特異的に誘導される遺伝子を検索した。3ラウンドのサブトラクション後に残ったcDNAをクローニングし、2000クローンの遺伝子配列を決定した。その結果、73個の既知の遺伝子と65個のESTおよび111個の未知の遺伝子が得られた。既知の遺伝子のうち4個はすでにE2Fの標的遺伝子として報告されているもので、サブトラクションが効率よく行われたと考えられた。得られた遺伝子が本当にE2Fによって発現誘導されるかどうかをノーザンブロッティングによって確認しているが、現在までのところ既知の遺伝子62個のうち19個が、EST21個のうち6個が、また未知の遺伝子49個のうち9個が、E2F1の過剰発現により発現誘導されることが確認されている。一方我々は、アフリカツメガエルの初期発生過程におけるMidblastula移行(MBT)にE2Fが重要な役割を果たしていることを明らかにした。そこで、MBTにおけるE2Fの標的遺伝子を明らかにするために、MBT前後の胚から抽出したmRNAを用いてサブトラクション法およびディファレンシャルディスプレイ法を行った。その結果、各方法によりMBT以後に発現が上昇する遺伝子の候補が150クローンおよび100クローン得られた。これらのクローンにつきノーザンブロッティングにより発現誘導を検定したところ、各方法によりMBT後に発現量が増加する遺伝子が一つずつ同定された。今後更にスクリーニングを続けるとともに、これらの新規E2F標的遺伝子のDNA複製の誘導における役割を検討していく予定である。
