G1期におけるDNA複製因子の発現と活性の制御機構の研究

研究概要

転写因子E2Fは、癌抑制遺伝子産物pRbの主な標的であり、細胞周期進行特にS期(DNA複製)の誘導に中心的な役割を果たす。そこでDNA複製誘導におけるE2Fの役割を調べるために、E2Fの新規標的遺伝子の探索とE2Fによる転写制御メカニズムの解析を行っている。まず休止期のラット繊維芽細胞株REF52にヒトE2F-1を過剰発現し、サブトラクション法を用いてE2F-1過剰発現細胞に特異的に誘導される遺伝子を検索した。2000クローンのcDNAをスクリーニングした結果、E2F-1によって発現誘導される遺伝子として、22個の既知の遺伝子と16個のESTまたはcDNAおよび3個の未知の遺伝子断片を同定した。プロモーター解析によりチェックポイント系およびDNA修復関連遺伝子であるClaspinおよびRad51, Rad51ap1, MLH1がE2Fの新規標的遺伝子であることを明らかにした。ESTおよび未知の遺伝子に関して、完全長cDNAのクローニングを行っている。またスクリーニングの過程で、外来性のE2Fによって発現誘導されるが細胞内のE2Fを活性化する血清刺激によっては発現誘導されない遺伝子を見いだした。E2Fの過剰発現によるアポトーシスの誘導に関与するp14^<ARF>も同様の制御を受けることを明らかにした。このことは、E2Fによる転写制御に新たなメカニズムが存在することを示している。そこで、p14^<ARF>のプロモーターを用いて、この新しい制御メカニズムの解析を行っている。一方、アフリカツメガエル初期発生過程において、E2FはMid Blastula移行(MBT)以後に重要な働きをすることを見いだした。そこでE2Fの新規標的遺伝子の探索を行う目的で、MBT以後に発現が上昇する遺伝子をサブトラクション法・ディファレンシャルディスプレイ法にて探索し、SSB1とProfilin、その他新規の候補遺伝子をいくつか同定した。今後、得られた新規E2F標的遺伝子の機能を解析していくとともに、E2Fによる新しい制御メカニズムの解析を行って予定である。
