G1期におけるDNA複製因子の発現と活性の制御機構の研究

研究概要

i)細胞周期のS期(DNA複製期)への進行に重要な転写因子E2Fの役割を調べるために、E2Fの新規標的遺伝子の探索とE2Fによる転写制御メカニズムの解析を行っている。既に同定している34個のE2Fの新規標的遺伝子のうちEST4個および未知の遺伝子断片3個に関して、完全長cDNAをクローニングした。これらの過程で、E2Fの過剰発現はp53の活性化に係わるp14ARFやCDKインヒビターp27Kip1等の増殖刺激で活性化されたE2Fには誘導されない遺伝子の発現を誘導することを明らかにした。また、E2Fの過剰発現は今までに報告のない異常な構造をもった転写産物を誘導することを明らかにした。以上から、E2Fには細胞周期に依存しない新たな転写制御メカニズムが存在することが示唆された。新規標的遺伝子の遺伝子発現を抑制するためのsiRNAの発現ベクターを作成したので、これらを用いてDNA複製誘導における新規標的遺伝子の役割を検討していく予定である。
ii)アフリカツメガエルにおいて、複製開始点におけるCDKインヒビターp27Xic1のユビキチン化と分解が複製開始複合体の活性化に必須であるため、哺乳類細胞におけるp27Kip1の制御機構を解析している。p27は複数のシグナル経路によって制御されるが、Jab1/COP9シグナロソームに注目し、Jab1のノックアウトマウスを作製し解析している。Jab1ヘテロマウスは正常に生まれるが、Jab1ヌルマウスは全く生まれてこなかった。Jab1ヘテロマウスは野生型のマウスに比べて体重の減少が見られ、p27ヌルマウスは野生型マウスに比べ巨大化することから、Jab1ヘテロマウスではp27の分解機構が破綻し個体の小型化がおこっていると推測された。今後は、p27と複製開始複合体の活性化の観点から、Jab1の遺伝子改変マウスのより詳細な解析を進める予定である。
