多様な生物におけるHis・Aspリン酸リレー型シグナル伝達の普遍的分子機構

研究概要	本研究計画に関して、本年度は高等植物(シロイヌナズナ)を対象として、生物時計機構に関わると思われる疑似リスポンスレギュレーター(APRR1・APRR3・APRR5・APRR7・APRR9)に関して主に遺伝学的な解析を行った。具体的には、APRR9、APRR7、及びAPRR5に関してそれぞれ機能欠損変異株を取得して、時計関連の表現型に関して詳細な解析を行った。APRR9、APRR7、APRR5の欠損変異株において、それぞれ時計機能に関する欠損が認められた。具体的には、APRR9欠損変異株においては概日リズムの周期が長くなっていた(あるいは位相が後退していた)。APRR7変異株においても概日リズムの周期が長くなったいたが、APRR5変異株においては逆に短周期になっていた。これらの現象は調べた限り全ての概日リズム制御遺伝子に関して確認された。これらの結果は、APRR9/APRR7/APRR5の各因子がそれぞれAPRR1(TOC1)と相補的な役割を担いながら時計中心振動体の近くで機能していることを強く示唆している。この考えは、APRR9がAPRR1と直接相互作用することが酵母two-hybrid法により示されたことからも支持された。また、APRR7やAPRR5の欠損変易株においては、光形態形成や開花時期制御に関しても顕著な表現型を示した。具体的には、これらの欠損変異株は胚軸伸長の測定において赤色光に対する感受性が低下していた。また、APRR7及びAPRR5両変異株において長日条件下での開花(花成)が顕著に遅延していた。本年度得られた結果は、前年度の成果として報告したこれら因子の過剰発現体の解析から得られた結果とも良く一致して、我々が新規に見いだした疑似リスポンスレギュレーター群が時計関連因子として重要な働きをしていることを強く示唆しているAPRR3に関しては解析が遅れているが、これは次年度の重要な課題である。これらの結果を総合することで、高等植物(シロイヌナズナ)における時計分子機構の本質に迫れることが大いに期待できる。
