His・Aspリン酸リレーによる大腸菌O157病原性遺伝子群の発現調節機構

研究概要

大腸菌におけるHis・Aspリン酸リレー情報伝達分子機構の解明を目的として、リン酸レギュロンの二成分制御系による遺伝子発現ネットワークの解明と、病原性大腸菌O-157特異的ゲノム解析を基にした二成分制御系による病原性遺伝子群発現の解析を目的とし、以下のことを明らかにした。
1)リン酸レギュロンにおけるモデュレーターPhoUの解析
リン酸レギュロン遺伝子の発現はPst S,A,B,C(リン酸の能動輸送系)およびモデュレーターPhoUによって間接的に負の調節を受けている。即ち高リン酸状態では、これら遺伝子産物はPhoRのもつフォスファターゼ活性を上昇させ、リン酸化型PhoBからのリン酸基離脱反応に関わる。結果的にPhoBの不活化が起きる。この機構がどのようなものであるか、今のところ十分な解析がなされていない。PhoUの機能を明らかにするため、ドミナントネガティブなphoU変異体を分離し変異部分を明らかにした。
2)腸管出血大腸菌O-157特異的二成分制御系の解析
O-157ゲノム長は5.6M塩基対(5,600kb)で、K12株と比べO-157特異的領域は約1.6M塩基対である。この特異的領域に存在する二成分制御系をホモロジーを基にコンピュータによる検索を行い、2組のO-157特異的二成分制御系を発見した。また、これら制御系により発現が制御される遺伝子領域を明らかにした。
