上皮細胞におけるアミノ酸・薬物のベクトル輸送形成機構の解析

研究概要

上皮細胞における薬物のベクトル輸送について、特に有機アニオン系化合物の胆汁排泄に焦点を当てた検討を進めた。極性細胞(MDCK細胞)にラット肝取り込みに関与するNtcp、ラット胆汁排泄に関与するBcrpを共発現させた。この細胞を介した8種類の胆汁酸の経細胞輸送を定量化した。MDCK細胞に内因性の存在するトランスポーターのため、taurocholateについてははNtcpを発現させるのみでベクトル輸送を達成しているが、BSEPの共発現により有意にベクトル輸送の効率は増加した。また、lithocholate以外の胆汁酸についてもNtcp/Bsepの共発現によりベクトル輸送を達成しており、本システムが胆汁酸の肝胆系輸送機構を明らかにする上で有用であることが明らかとなった。
ヘテロダイマー型アミノ酸トランスポーターの補助サブユニットに内在すると考えられる管腔側膜あるいは血管側膜へのsortingシグナルを明らかにするために、2つの補助サブユニット間の分子キメラを作製し、MDCKII細胞に発現させた。活性サブユニットの膜移行を観察することにより、補助サブユニットのN-末端領域にsortingに関わる領域が存在することが明らかになった。また、活性サブユニットBAT1の日本人型シスチン尿症変異(P482L)の解析により、細胞膜移行における活性サブユニットのC-末端の重要性を明らかにした。この部位に結合する複数個のタンパク質を2次元電気泳動により単離し、タンパク質特定のための解析を進行させている。
