研究概要	本研究では、上皮細胞等におけるイオン・水の極性輸送(ベクトル輸送)の分子基盤を解明し、その異常により生ずる疾患の病態理解や治療法の開発を目的としている。特に内向き整流カリウムチャネルKir4.1・Kir5.1、水チャネルAQP4を対象とし、細胞内局在機構とチャネル機能の理解を目指している。Kir4.1は、腎尿細管上皮細胞では基底側膜にてK^+のrecyclingに携わっているが、更に胃壁細胞の頂上膜にてH^+-K^+ATPaseと共役し、胃酸分泌に関与している知見を得た。また、Kir4.1とKir5.1は、脳・網膜のグリア細胞や腎尿細管では一部ヘテロ複合体を作っているが、内耳では各々ホモ複合体として存在し機能していることを見出しつつある。特に網膜ミュラー細胞では、Kir5.1はendfoot・血管周囲の突起には存在しないが、シナプス周囲の突起にKir4.1と共発現していた。これは、神経活動により上昇したK^+はKir4.1-Kir5.1ヘテロ複合体を通してグリア細胞へ吸収され、Kir4.1ホモ複合体を通して放出されている事を示唆する。従って、Kir4.1、Kir5.1のホモ、ヘテロ複合体は各々異なる機能を持ち、更には異なる細胞内局在制御を受け、各組織・細胞内部位にて特有の役割を果たしていると考えられた。Kir5.1は単独で機能的チャネルを構成しないが、アンカー蛋白PSD-95と共発現すると、チャネル活性を持つことを見出した。Kir5.1のC末端部のセリンがPKAリン酸化されると、この両者の結合が解離しチャネル活性が消失する知見も得た。Kir4.1とAQP4の上皮での局在機構をMDCK細胞で解析した。Kir4.1とAQP4は共に基底外側膜へ分布し、Kir4.1はC末端のPDZドメイン結合モチーフ(SXV)が局在に重要であったが、AQP4ではC末端26アミノ酸が必要であった。
