イオン・水ベクトル輸送におけるカリウムチャネル極性分布の役割とその分子基盤

研究概要	本研究では、上皮細胞・グリア細胞等におけるイオン・水の極性輸送(ベクトル輸送)の分子基盤を解明し、その異常により生ずる疾患の病態理解や治療法の開発を目的としている。特に内向き整流カリウムチャネルKir4.1・Kir5.1、水チャネルAQP4を対象とし、細胞内局在機構とチャネル機能の理解を目指している。以前に我々は、Kir4.1か、胃の壁細胞や内耳の血管状といった上皮組織や、脳および網膜のグリア細胞に分布し、K^+のベクトル輸送に深く関わっていることを見出していた。最近の研究で更に、(1)腎尿細管上皮細胞では、Kir4.1はKir5.1とヘテロ複合体を作って基底側膜に分布し、K^+のリサイクリングに携わっていること、(2)網膜のグリア細胞(ミュラー細胞)では、Kir4.1/Kir5.1のヘテロ複合体がシナプス周囲の突起に、Kir4.1のホモ複合体がendfoot・血管周囲の突起に分布しており、前者は神経活動により上昇した細胞外のK^+の吸収を担い、後考は回収したK^+を硝子体や血管腔へ放出する機能を果たしている可能性があること、(3)内耳において、Kir4.1,Kir5.1はホモ複合体として蝸牛側壁の異なった場所に存在し、K^+の循環に対してそれぞれ正、負の制御を与えている可能性、を見出した。また、脳のアストロサイトでは、血管周囲の突起にKir4.1/Kir5.1のヘテロ複合体、神経周囲の突起にヘテロ複合体とKir4.1のホモ複合体の両者が発現しており、ミュラー細胞とは違った分布を示していることも明らかとなりつつある。従って、Kir4.1、Kir5.1のホモ、ヘテロ複合体は、各組織・細胞内部位において異なる細胞内局在制御を受けることで特有の局在パターンを示していることが示唆された。更にはそれらチャネルの局在が、細胞・組織特異的な生理的機能に貢献していることも考えられた。
