研究概要

タイトジャンクションの機能に関わるクローディン1の役割を解明する目的で、遺伝子標的組換えの技術によりクローデイン1遺伝子を欠失させた変異マウスを作製したところ、ホモに変異を持つマウスは出生後24時間以内に死亡することが明らかになった。これらのマウスでは皮膚に皺を呈するという異常が見られたため、正常マウスとクローディン1欠失マウスの皮膚におけるタイトジャンクションの局在とその機能について重点的に比較検討した。その結果、表皮細胞にクローデイン1が非常に強く発現しており、しかも顆粒細胞に連続したタイトジャンクションが存在することが明らかになった。さらに、深部組織から表皮へのマーカーの拡散度を調べたところ、正常マウスでは顆粒細胞層に局在するタイトジャンクションの位置でマーカーの拡散が留まることが分かった。すなわち、ホモ変異マウスでは皮膚からの水分蒸散量が野生型、ヘテロマウスに比べて著しく昂進しており、それに伴う脱水によりマウスが死に至ると考えられた。以上の結果は、哺乳類の皮膚のタイトジャンクションが水分蒸散を防ぐ皮膚のバリア機能に必須であることを意味し、皮膚のバリアにタイトジャンクションは関与しないというこれまでの通説を覆すものである。
