研究概要	オートファジーはダイナミックな膜動態を伴うリソソームによるタンパク質分解機構で、飢餓条件で著明に誘導される。哺乳類では、細胞の内外の環境変化に速やかに応答して不要な、あるいは障害オルガネラやタンパク質を処理して恒常性を維持するのに働いている。本研究では、高等動物におけるオートファジーの生理機能、病理機能を明らかにするため、オートファジーに必須な遺伝子群Apgシステムの同定および機能解析を進めている。ヒトAPG遺伝子群の中、とくにユビキチン類似の蛋白修飾システムであるApgコンジュゲーションシステムを構成する遺伝子群に着目している。昨年度までにApg12をモディファーアーとするシステムとApg8をモディファーアーとするシステムが協調的に働き、オートファゴソーム膜の形成に関与していることが明らかになってきた。このシステムの中核にあるApg7というE1様酵素に続いて、ヒトhApg3pホモログの単離・解析をおこない、hApg3pが、Apg8ホモログであるMAP-LC3,GABARAP,GATE-16を基質としたE2酵素であることを証明した。hApg3pはApg7およびApg12とも相互作用があり、相互作用の実体とMAP-LC3,GABARAP,GATE-16のオートファゴソーム膜への移行との関係を現在検討している。また、組織、細胞におけるApgコンジュゲーションシステムの分布を調べたところ、驚いたことにオートファジーが盛んでない細胞にこのシステムが大量に存在することを見出した。そこで、視点を拡げ、これら2つのコンジュゲーションシステムがオートファジー以外の膜形成を伴う様々な細胞機能(ファゴサイトーシス、メラノゾーム形成など)にも、働いていることを解析中である。
