研究概要	オートファジーはダイナミックな膜動態を伴うリソソームによるタンパク質分解機構で、飢餓条件で著明に誘導される。哺乳類では、細胞の内外の環境変化に速やかに応答して不要な、あるいは障害オルガネラやタンパク質を処理して恒常性を維持するのに働いている。本研究では、高等動物におけるオートファジーの生理機能、病理機能を明らかにするため、オートファジーに必須な遺伝子群Apgシステムの同定および機能解析を進めている。ヒトAPG遺伝子群の中、とくにユビキチン類似の蛋白修飾システムであるApg合修飾システムを構成する遺伝子群に着目している。Apg8ホモログの一つLC3は、Apg7とApg3の修飾を受けてオートファゴゾーム膜に結合する。このプロセスには、Apg12結合修飾システムとLC3結合修飾システム相互の正・負の制御が関与していることを明らかにした。LC3,とGABARAP(もう一つのApg8ホモログ)が膜結合型のII型となる際、酵母のようにlipidationを受けていることを証明したが、ホスホエタノールアミン(PE)と結合しているかどうかは不明で、検討中である。細胞内のApg12、LC3プールは小さく、ユビキチンシステムと同様、脱Apg化反応が生体の要求に応じて作動することがこの結合修飾を保証する。これを保証する脱Apg化反応に、Apg4Bが関与する示唆を得た。Apgシステムの生理的役割では、腎臓podcyteでは、オートファジーは細胞分化に関与する示唆を得た。個体レベルでのApgシステムAPG7の役割を調べるため、コンディショナルノックアウトマウスを作製した.Cre発現アデノウィルスを用いてAPG7を肝臓で欠損させ,その表現型を解析している.またAPG7欠損マウスも作成中である。
