研究概要	環境中の金属濃度の変動に直接接触している大腸菌が、細胞内の金属濃度を一定に維持するホメオステーシス制御に関与する遺伝子群の発現制御機構の全体像の解明を目指した研究を実施し、次の成果を得た。 1)大腸菌には外界の9種類の金属の濃度を感知し応答する転写因子が少なくとも12種類存在する。それら転写因子の大量発現系を構築し、全ての純化に成功した。 2)金属応答転写因子支配下の遺伝子群をDNAチィップを利用したマイクロアレイ解析で同定し、そのプロモーターの転写シグナルとしての活性と制御を解析した。 3)金属応答転写因子支配下遺伝子のプロモーターを単離し、試験管内転写反応で転写因子作用様式を系統的に解析した。 4)転写因子との相互作用によるRNAポリメラーゼの機能特異性変化機構が、転写因子の接点に相関するとの我々の理論を証明するために、蛋白-蛋白相互作用接点同定を目的に人工プロテアーゼFeBABEを利用した接点依存的切断法を開発した。 5)金属応答転写因子の抗体を作製し、それら転写因子の大腸菌内濃度の、外界金属濃度変化に相関した変化を系統的に観測した。
