脊索動物モデルのホヤ発生の遺伝子システムとその進化の解明

研究概要	脊索動物門はホヤなどの尾索類、ナメクジウオの頭索類、それに脊椎動物からなる。これらの動物は脊索・背側神経管・鰓裂など多くの共有形質を有し、共通の祖先から進化してきたものと考えられている。ホヤのオタマジャクシ幼生は体幹部背側の中枢神経系(構成細胞数約350)、尾部の脊索(構成細胞数40)や筋肉(構成細胞数約40)など、脊椎動物の体制の原型がそこに存在する。本研究は、カタユウレイボヤ(Ciona intestinalis)のcDNAプロジェクトおよびゲノムプロジェクトの展開を計り、発生遺伝子システムの解明と進化と関連したゲノム科学の進展を計ることを目的とした。まずcDNAプロジェクトとして、この動物の発生および成体で発現する遺伝子のEST解析を行い、これまでに480,000近いEST情報を得た。これらのESTを整理したところ、約18,000のユニークなcDNAクローンであることがわかり、これを再配列した"Ciona Gene Collection"を作り、世界中の研究者に配布している。また、現在、カタユウレイボヤ発生における各遺伝子の時間的・空間的発現パターンの網羅的理解を求めて解析中である。次にゲノムプロジェクトでは、アメリカJGI、国立遺伝研(日本)と共同で、カタユウレイボヤのドラフト・ゲノムを決定した(2002年末)。その結果、この動物のゲノムサイズは153-159Mbp、タンパク質をコードする遺伝子は15,852と見積もられた。本年度は、このゲノムおよびcDNA情報をもとに、この動物の発生関連遺伝子のできるだけ正確なアノテーションを進め、公表した。これらの結果は、これからこの動物の発生遺伝子を研究するための大きな基盤となると考えられる。
