モデル多細胞生物発生の遺伝子システムの全体像解明と計算機モデル化

研究概要	発生の遺伝子システムの全貌解明を何らかの系でやりとげるため、受精卵から成虫(959体細胞)の全細胞系譜がわかっており、全ゲノムDNA配列が多細胞生物で最初に決定された線虫C.elegansを用いて、構造、発現、機能、進化研究をいわば四位一体で進め、この結果を逐次統合し、発生の遺伝子システムの解明をおこなうことを目的とした。さらには発生の計算機モデル化をめざした。 1)cDNAプロジェクトから分類同定した約1万種のcDNAのうち9000種についてwhole mount in situハイブリダイゼーションによる発現パターン解析を終えた。これらはNEXTDB<http://nematode.lab.nig.ac.jp>で公開した。発現パターンのアノテーションづけを詳細におこなった。 2)発現パターンに基づくRNAi解析を進めた。母性発現遺伝子のうち、「2細胞期から原腸陥入開始期までにその発現が消失あるいは局在する遺伝子群」に注目した結果、65%という高率で表現型が見られ、重要遺伝子が濃縮されたことを示した。強いF1胚致死を示す61遺伝子について4D顕微鏡で詳細に調べた結果、受精/最終減数分裂から8細胞期までの時系列での異常に分類できた。そのうち最も早期の異常である「受精後分裂なし」は主としてプロテアソーム遺伝子であった。 3)発現パターンのクラスタリング解析をおこなった。約7000遺伝子のデータは因子分析で時間軸と空間軸にそって54クラスターに分類され、さらにウォード法で詳細なクラスタリング解析をおこなった。これらの遺伝子群について上流配列を切り出し、共通モチーフ探索をおこなった。 4)Diploscapter sp.について4D解析を行った結果、4細胞期に1直線上に並ぶことを見いだした。C.elegansでは配置異常に相当するものである。 5)細胞の形状を力学モデルによって構成し,細胞分裂にともない細胞配置が決定される現象を再現するシミュレータを構築した。
