モデル多細胞生物発生の遺伝子システムの全体像解明と計算機モデル化

研究概要

発生の遺伝子システムの全貌解明のため、受精卵から成虫(959体細胞)の全細胞系譜と全ゲノムDNA配列が明らかにされている線虫C. elegansを用いて、構造、発現、機能、進化研究をいわば四位一体で進めた。
1)EST配列12万本をゲノム配列と,比較し、約1万遺伝子のエキソン・イントロン構造を確定した。選択的スプライシングの結果、遺伝子あたり平均1.7のmRNA種、1.4のタンパク質種ができることを見いだした。また、新たなスプライスリーダーをSL12まで8種見つけた。分類同定した約1万種のcDNAについてwhole mount in situハイブリダイゼーションによる発現パターン解析をおこない、NEXTDB<http://nematode.lab.nig.ac.jp>で公開した。発現パターンのアノテーションづけを詳細におこない、発現パターンの類似度の検索エンジン、SearchEXを開発した。
2)発現パターンのクラスタリング解析をおこない、同一クラスタ遺伝子の5'上流にモチーフ候補を見いだした。
3)母性mRNAの翻訳制御について、pos-1、pip-1/spn-4などの複数因子の組み合わせによるfine tuningの機構を明らかにした。
4)Diploscapter sp.について完全長cDNAライブラリーを構築し、比較発現解析を開始した。
5)細胞の形状を力学モデルによって構成し,細胞分裂にともない細胞配置が決定される現象を再現するシミュレータを構築し、8細胞期までの解析をおこなった。
6)ゼブラフィッシュの皮膚における色素細胞の存在様態の詳しい解析をおこない、melanophoreとxanthophoreの2種類の色素が共存している細胞の存在を確認した。
7)皮膚に縞模様が形成される変異マウス(TWマウス)のパターン形成原理を解析した。
8)鳥の羽にはいろいろなパターンがあるが、反応拡散の原理から説明できることを示した。
