研究概要	(1)モデル多細胞生物発生の遺伝子システムの全体像解明と計算機モデル化(担当:小原) 1)EST配列12万本からエキソン・イントロン構造を確定した約1万遺伝子についてwhole mount in situハイブリダイゼーションによる発現パターン解析をおこない、NEXTDB<http://nematode.lab.nig.ac.jp>で公開し、これをもとにした共同研究を世界中で進めた。発現パターンのクラスタリング解析をおこない、同一クラスタ遺伝子の5'上流にモチーフ候補を見いだし、実験的検証を進めた。 2)glp-1母性mRNAの翻訳制御メカニズムについて、pos-1、pip-1/spn-4などの複数遺伝子の組み合わせによるfine tuningの機構を明らかにし、3'-UTRの結合配列を明らかにした。 3)近縁線虫Diploscapter sp.についてcDNAライブラリーを構築し、EST約2万本から約6600種に分類でき、C.elegans,C.briggsaeとのオルソログリストを作成した。スプライスリーダー配列はC.elegansのものと同じであった。発現パターンについてC.elegansとの比較を開始した。 4)細胞の形状を力学モデルによって構成した胚発生シミュレータ(8細胞期まで)を構築し、中心体の動きなどをより正確に再現するような条件を求めた。 (2)生物発生のコンピュータシミュレーション(担当:近藤滋) 1)レーザー除去実験による色素細胞相互作用の同定をおこない、パターン形成に関与しているのは、黒色・黄色の2種類の色素細胞であること、それらの相互作用がチューリングの関係を満たすことなどが明らかにした。 2)Foxn1遺伝子に変異のある新しいアレルのマウスの皮膚に、BZ波と同じパターン変化する移動するストライプ模様がおきることを発見し、移動波が皮膚模様形成を起こす例を初めて示した。
