気管支喘息・アトピー、精神分裂病、血清脂質異常関連遺伝子の解明

研究概要	喘息・アトピー、精神分裂病、血清脂質異常,急性心筋梗塞の関連遺伝子を全ゲノムにわたって検索し、遺伝子を同定することを目的とした。気管支喘息ではこれまでの申請者らによる研究で連鎖が見られた5番染色体長腕に存在する遺伝子の変異検索を行い、内部対照関連解析を行うことにより、喘息遺伝子の同定を目指した。5q31-q35の範囲にある遺伝子の中で連鎖の最も強い領域の48遺伝子について解析を行った。この結果、IL4遺伝子およびそれに近接するIL13遺伝子の変異と気管支喘息との有意な関連を検出し、その他にSPINK5遺伝子およびHMMR遺伝子の変異とアトピー性皮膚炎との有意な関連を検出した。5番染色体以外では、4番染色体にあるH-PGDS遺伝子と気管支喘息、IRF2遺伝子とアトピー性皮膚炎、6番染色体にあるPAFAH遺伝子と気管支喘息、12番染色体にあるAID遺伝子と気管支喘息との関連をそれぞれ検出した。精神分裂病は罹患同胞対法により日本人の精神分裂病との連鎖解析を行い、22qおよび3pに連鎖を示唆する領域が検出された。さらに、これらの連鎖領域に存在する遺伝子の変異検索を行い、3pに存在しているCHL1遺伝子と精神分裂病との関連を検出した。眼球運動を利用した精神分裂病のQTL連鎖解析により、22qに連鎖を検出し、罹患同胞対法と同様の結果が得られ、日本人における精神分裂病の疾患感受性遺伝子が22qに存在する可能性が強く示唆された。血清脂質異常では、血清総コレステロール、HLDコレステロール、中性脂肪について全ゲノムにわたる同胞対のQTL解析を行い、3qに連鎖領域を検出した。
