研究概要	慢性関節リウマチ(RA)、全身性エリテマトーデス(SLE)の病因・病態を解明するために、本研究では、病態上の重要性、mRNA発現プロファイリング、連鎖解析、動物モデルの情報などに基づいて設定した多数の候補遺伝子の変異解析と関連分析により、疾患感受性遺伝子を検出することを目的とした。今年度は、以下の成果が得られた。 (1)RA滑膜に発現が増強する遺伝子をdifferential display法を用いて検索し、滑膜におけるId family遺伝子の発現を検出した。Idはfibroblast増殖、血管新生誘導因子として働くことが知られているため、BAの病態に深く関連する可能性が考えられ、さらに、その遺伝子群は染色体上のRA候補領域に位置するため、疾患感受性遺伝子としての可能性も考えられる。本年度は、これらのうち、1p36に位置するID3の変異解析を施行し、2個所の単一塩基多型(SNP)と、非同義置換をコードする1個所の稀な変異を検出した。 (2)FcγRIIBはB細胞・単球において抑制性シグナルを伝達することが知られ、また、この遺伝子が位置する1q23領域はSLEの候補領域であることから、FCGR2Bを有力な候補遺伝子と考え、変異解析を行い、cDNAを用いたnested PCRにより翻訳領域にアミノ酸置換を伴う多型を検出し、SLEと有意に関連することを示唆する結果を得た。 (3)B細胞のシグナル伝達分子であり、マウスモデルにおいて自己免疫との関連が強く示唆されているCD19を候補遺伝子と考え、変異スクリーニングを施行した。多数のSNPに加え、3'非翻訳領域(UTR)にGTリピート多型が見出された。うち、翻訳領域の同義置換、イントロンのSNP、3'UTRの反復配列多型とSLEとの有意な関連を検出した。 (4)このほか、SLE,自己免疫との関係が近年特に注目されるBCMA,NKG2Aの多型解析を行い、多数の新規の多型を検出したが、RA,SLEとの関連は検出されなかった。
