遺伝子発現を基盤とする病態機序の解明法の開発

研究概要	遺伝子発現のモデル化:組織内でのmRNAの組成がZipfの法則に従うという観察に対して、各遺伝子の発現量が点突然変異によって変動し、かつその変動量がもとの発現量と一定の確率関数の積で与えられるものであるとすることで説明できることを証明した。発現データ統合:公的に大量に存在する遺伝子発現データから正常解剖学的発現パタンデータを選択しEST・SAGE・マイクロアレイ・iAFLPについてサンプルの違いを生物学的な解釈で吸収し意味のある比較を行った。その結果公的に存在する発現データはそのカバー率や精度(相互一致度)において予想外に悪く、ヒト遺伝子の発現量が少ない半数についてはデータが不在であると表現できることが判明した。病態解析:乾癬・シェグレン症候群の患者サンプルRNAのiAFLPによる測定値から、疾患特異的発現様式を同定した。遺伝子機能のエンコード:遺伝子機能の言語表現を用いて遺伝子の機能を各種の用語や教科書中のページとの関係として表現するシステムを前年度までに作成していたが、教科書インデックスデータの統合・インデックスデータ行列の特異値分解による次元下げ近似・教科書用語辞書を使った任意の文書のインデックス付け・任意の文書(偽ページベクトル)の教科書空間へのマッピングの4工程を自動的に行うシステムを構築した。4万弱の教科書用語は手作業による意味分類を行い、用語辞書の付加価値を高めた。また辞書用語による任意の文書のインデックス化の結果を解析し、文書中のほかの用語に比べ蛋白や遺伝子名が特別に表記のバリエーションが多くその重要度に反して自動インデックス付けの効率が極めて低いことを発見し、今後の辞書の拡張やマッピングの方法に対する重要な改善指針を得た。
