糖尿病関連遺伝子群の解明

研究概要	糖尿病の発症予防をする上で、その感受性遺伝子の同定が必須である。本研究は、single nucleotide polymorphism(SNP)を用いた候補遺伝子解析等による糖尿病感受性遺伝子の同定を目的とする。更に我々がその遺伝子発現がインスリン抵抗性に関係することを明らかにしたphosphodiesterase 3B(PDE3B)について、その活性化制御因子をクローニングし、糖尿病との関連を解析する。本年度、倫理面では、3省新指針準拠の計画書が愛媛大学医学部倫理委員会で8月に承認され、これまで対照者650名、2型糖尿病300名、1型糖尿病200名のDNAサンプルを収集した。糖尿病患者5000名について1型糖尿病マーカーのglutamic acid decarboxylase(GAD)抗体のスクリーニングを行い、陽性率(4%)を決定した。また、各候補遺伝子について全エクソンシークエンスによりSNPを同定した後に、連鎖不平衡とハプロタイプ頻度を検討し、糖尿病との関連を解析した。1型の新候補auotoimmune regulatorに新たなミスセンス変異3種を同定した。2型については、インスリンで4種、インスリン受容体遺伝子で9種、新候補のresistinで3種のSNPを同定したが、糖尿病とは関連しなかった。また、インスリン受容体異常症2例に新変異を同定した。更に、脂質代謝関連遺伝子のSNPの組み合わせを解析したが、2型糖尿病との関連を認めなかった。一方、PDE3Bと相互作用する蛋白Xを酵母two-hybrid法によりクローニングし、脂肪細胞内でインスリンが相互作用を増強することを証明した。XにSNP3種を同定した。また、マウスとヒトPDE3Bプロモーターをクローニングし転写開始点を決定した。マウスプロモーター活性が脂肪分化により増強することを証明し、ヒトではSNP4種を同定した。
