研究概要

糖尿病発症予防法の確立には感受性遺伝子の同定が必須である。本研究は、single nucleotide polymorphism(SNP)を用いた候補遺伝子解析等による同定を目的とする。更に、申請者らがその遺伝子発現とインスリン抵抗性との関係を明らかにしたphosphodiesterase 3B(PDE3B)について、活性化制御因子をクローニングし、糖尿病との関連を解析する。
倫理面では、3省新指針準拠の計画書が平成13年に承認され、これまでに対照750名、2型糖尿病450名、1型糖尿病168名のDNAを収集した。全ゲノム解析用に対照と2型各20名を送付した。
1型糖尿病マーカーのglutamic acid decarboxylase(GAD)抗体を糖尿病5000名でスクリーニングした。GAD陽性をインスリン依存(IDDM)と非依存(NIDDM)に分類し、HLAを解析した結果、1型感受性ハプロタイプ3種のうち、DRB1*0405-DQB1*0401のみがGAD陽性NIDDMと関連した。
各候補遺伝子についてSNP及びそれによるハプロタイプについて糖尿病との関連を解析した。1型の候補auotoimmune regulatorに高頻度のSNP5種とミスセンス変異3種とを同定した。2型の候補では、resistinでSNP3種を同定したが、2型とは関連しなかった。FOXC2では、高頻度SNP3種と主要ハプロタイプ4種は2型と関連しなかったが、1種のミスセンスLeu303Met(+907C>A)は2型で多い傾向を認めた。
PDE3Bと相互作用する14-3-3蛋白を酵母two-hybrid法によりクローニングし、脂肪細胞内でインスリンが相互作用を増強することを証明した。また、ヒトPDE3BプロモーターにおいてSNP4種とinsertion1種を同定した。
